Allocation du carbone et métabolisme azoté chez l'haptophyte Tisochrysis lutea ArchiMer

The aim of this thesis is to improve knowledge on mechanisms involved in the response to nitrogen limitation and in lipid accumulation in the microalgae haptophyte Tisochrysis lutea. The wild type strain and a lipid accumulating mutant strain were grown under different nitrogen limitation and starvation and analyzed by functional genomics. Four genes of high-affinity nitrate/nitrite transporter (Nrt2) were identified and characterized to reveal the mechanisms involved in mineral absorption in this species. Transcriptomes of both strains were sequenced and proteins affected by nitrogen starvation and differentially expressed between the two strains were identified. Caractérisation morphologique et physiologique des microalgues utilisées pour la nutrition larvaire de l'huître creuse, Crassostrea gigas ArchiMer

En aquaculture, sur une cinquantaine d'algues testées, seulement une douzaine présente un réel intérêt en écloserie-nurserie de bivalves (Chrétiennot-Dinet et al, 1986). Le premier but de notre travail est de caractériser 11 espèces de microalgues au cytomètre en flux afin de fournir un outil descriptif, facilement réutilisable, de ces espèces et d' évaluer l' influence de l'état physiologique et de la méthode de fixation sur les signatures cytométriques. Les paramètres mesurés au cytomètre en flux sont la FSC, la FL3 et la SSC liées respectivement à la taille, la fluore scence et la structure des cellules phytoplanctoniques. Microalgae are at the basis of aquatic food webs and may be the indirect target of herbicides from agricultural and urban uses. They also interact with other compounds from their environment such as natural dissolved organic matter (DOM), which can itself interact with herbicides. This thesis aimed to study, in laboratory controlled conditions, the influence of natural DOM on herbicide toxicity to microalgae by using monospecific, non-axenic cultures. Interactions microalgues-bactéries en système expérimental bispécifique : effets sur la croissance de Dunaliella sp. ArchiMer

The use of algae-bacteria mixed culture has been suggested to improve performance of industrial microalgae culture. The present study aims to investigate impact of selected bacteria on Dunaliella sp. growth. 97 bacteria strains were isolated from a variety of marine microalgae cultures. The diversity of strains revealed members of α-and γ-proteobacteria, Cytophaga-Flavobacterium-Bacteroides and grampositive. A two-phase approach was then implemented to select bacteria that improve Dunaliella sp. growth, using an original high-throughput experimental device. Nowadays, spat production of Pacific oyster Crassostrea gigas relies on rearing techniques under controlled conditions in hatchery. In this context, an extensive research in aquaculture is necessary because larval development and metamorphosis are critical for hatchery management. La synthèse des informations collectées en 1988 en Charente Maritime, en matière de développement microalgaux a pour but de mettre en évidence, à partir des données recueillies au cours d'une année, des tendances et niveaux de contaminations phytoplanctoniques, ainsi qu'une certaine identité du secteur surveillé. L'année 1988 est marquée par une efflorescence monospécifique de Gymnodinium sp. en période estivale et par l'apparition du Dinophysis à des concentrations supérieures au seuil toxique sans qu'aucne toxicité ne soit décelée. Test écotoxicologique sur microalgues et lecteur de plaques : mesures d'activités enzymatiques sur cellules in vivo. Application à l'évaluation de la qualité du milieu ArchiMer

A new method for the assessment of several contaminant effects on marine microalgae (Tetraselmis suesica, Skeletonema costatum) has been developped. The method is based on the measure of cells esterase activity using a fluorimetric stain, fluorescein diacetate (FDA). The experiments are performed in microplates using a fluorimetric microplate reader. The different steps of the experiments and the data processing are controlled by an original specific software. Application of the system for rapid ecotoxicological tests (determination of LC 50) and for assessment of environment quality by observation of water samples are proposed.

The food quality of Tetraselmis suecica slurry for Crassostrea gigas spat ArchiMer

In an attempt to provide cost-effective alternatives to live microalgae and to simplify hatchery-nursery procedures, concentrated microalgae appear to be very promising. Due to its high preservation ability, Prasinophytes appear to be good candidates for such alternative diets. Nevertheless, in a previous study the utilization of Tetraselmis suecica was only conceivable for old Crassostrea gigas larvae. In the present work its nutritional quality has been accordingly searched on ]apanese oyster's juveniles. Whatever the assemblages and condition, fresh or preserved microalgae, T. suecica did not allow an adequate postlarval development in C. gigas.

In an attempt to simplify mollusc hatchery-nursery procedures and to provide cost-effective alternative diets, the use of concentrated and preserved microalgae is an alternative to the use of fresh microalgae. Because of their ability for preservation, Prasinophytes appears to be good candidates for remote algal production. In the present work, the food value of fresh and preserved Tetraselmis chui and T. striata for umboned Crassostrea gigas larvae was studied. During the first experiment, fresh T. striata led to the best larval development, but these performances were not confirmed subsequently.
